The main objective of the game is to run away from a massive flying polyp, which will capture the players, resulting in the game's end. Players must defend themselves from enemies' attacks while proceeding as far as possible. In this game, players must be careful and dodge traps alongside fighting their enemies. By shooting their weapons at enemies, players can protect themselves.

An essential feature of Lovecraft’s Mythos Run is that players can unlock various heroes, including Detective, Professor, and Witch, as they progress through the game.
